    AI helps Stanford scientists discover “natural Ozempic” without the usual side effects

    Scientists have identified a natural molecule that produced Ozempic-like appetite suppression and fat loss in animals without several of the drug’s common side effects. Credit: Shutterstock

    Stanford Medicine researchers have identified a naturally occurring molecule that may suppress appetite and reduce body weight in a way that resembles semaglutide, the active ingredient in Ozempic. In animal studies, the molecule also appeared to avoid several problems associated with the drug, including nausea, constipation and substantial muscle loss.

    The molecule, known as BRP, works through a different but related metabolic pathway and activates a separate group of neurons in the brain. That distinction could make it a more precise tool for controlling appetite and body weight.

    A More Targeted Approach to Appetite Control

    “The receptors targeted by semaglutide are found in the brain but also in the gut, pancreas and other tissues,” said assistant professor of pathology Katrin Svensson, PhD. “That’s why Ozempic has widespread effects including slowing the movement of food through the digestive tract and lowering blood sugar levels. In contrast, BRP appears to act specifically in the hypothalamus, which controls appetite and metabolism.”

    The hypothalamus is a small region deep within the brain that helps regulate hunger, body temperature, hormone activity and energy use. Because BRP appears to act mainly in this area, it may influence appetite without producing as many effects elsewhere in the body.

    Svensson has co-founded a company that plans to begin clinical trials of the molecule in humans in the near future.

    Svensson is the senior author of the research, which was published March 5 in Nature. Senior research scientist Laetitia Coassolo, PhD, is the lead author of the study.

    Artificial Intelligence Reveals Hidden Peptides

    The discovery depended heavily on artificial intelligence, which allowed the researchers to search through proteins belonging to a group known as prohormones.

    Prohormones are inactive precursor molecules. They do not perform their final biological function until enzymes cut them into smaller fragments called peptides. Some of these peptides then act as hormones, carrying signals that influence metabolism, appetite and other complex processes in the brain and throughout the body.

    A single prohormone can be cut in several different ways, producing many possible peptides. Finding the biologically important ones is difficult because genuine peptide hormones are relatively rare and can be buried among large numbers of ordinary fragments created during normal protein processing and breakdown.

    Traditional laboratory methods can isolate and identify peptides, but the process can produce enormous amounts of data. Researchers may need to sort through hundreds of thousands of molecules to find the few that have meaningful effects.

    Searching for New Metabolic Signals

    The team concentrated on an enzyme called prohormone convertase 1/3. This enzyme cuts prohormones at specific amino acid sequences and has previously been linked to obesity in humans.

    One of the peptides produced through this process is glucagon-like peptide 1, or GLP-1. GLP-1 helps regulate hunger and blood sugar, and semaglutide works by copying its effects in the body.

    The researchers reasoned that the same enzyme might produce other peptides that influence energy balance and appetite. To find them, they turned to artificial intelligence.

    Peptide Predictor

    Rather than manually extracting proteins and peptides from tissues and then using methods such as mass spectrometry to identify huge numbers of molecules, the researchers created a computer algorithm called Peptide Predictor.

    The program searched all 20,000 human protein-coding genes for the types of sites where prohormone convertases typically cut proteins. The researchers then narrowed the search to genes that produce proteins secreted outside the cell, a common feature of hormones, and that contained at least four possible cleavage sites.

    That process reduced the field to 373 prohormones, giving the team a much more manageable group to investigate.

    “The algorithm was absolutely key to our findings,” Svensson said.

    Peptide Predictor estimated that prohormone convertase 1/3 could produce 2,683 distinct peptides from those 373 proteins. Coassolo and Svensson then focused on sequences that seemed most likely to affect the brain.

    They selected 100 peptides, including GLP-1, and tested whether they could stimulate neuron-like cells grown in the laboratory.

    A Tiny Peptide With an Outsized Effect

    As expected, GLP-1 strongly activated the neuronal cells, increasing their activity to three times the level seen in untreated control cells.

    One much smaller peptide produced an even more dramatic response. Made from only 12 amino acids, it increased neuronal activity tenfold compared with controls.

    The researchers named the peptide BRP after its parent prohormone, BPM/retinoic acid inducible neural specific 2, or BRINP2 (BRINP2-related-peptide).

    Amino acids are the basic building blocks of proteins and peptides. A molecule containing only 12 of them is extremely small compared with most full-sized proteins, yet BRP produced the strongest response in the initial cell tests.

    Food Intake Fell by Up to 50%

    The researchers next tested BRP in lean mice and minipigs (which more closely mirror human metabolism and eating patterns than mice do).

    An intramuscular injection given before feeding reduced food intake during the following hour by as much as 50% in both species.

    The team also gave daily BRP injections to obese mice for 14 days. On average, the treated animals lost 3 grams, with nearly all of the reduction coming from body fat. Mice in the control group gained about 3 grams over the same period.

    The treated mice also showed improved glucose and insulin tolerance. These measures reflect how effectively the body regulates blood sugar and responds to insulin, the hormone that helps move glucose from the bloodstream into cells.

    No Clear Signs of Common Side Effects

    Behavioral testing found no meaningful differences between treated and untreated animals in movement, water consumption, anxiety-like behavior, or fecal production.

    The absence of changes in fecal production was especially notable because semaglutide can slow digestion and cause constipation. The researchers also did not observe the nausea-related responses or major muscle loss associated with some existing weight loss treatments.

    Additional measurements of brain activity and body function showed that BRP acts through metabolic and neuronal pathways that differ from those activated by GLP-1 or semaglutide.

    Those findings suggest that BRP may reduce appetite through a more focused biological route, although the results remain limited to animals.

    Questions Before Human Testing

    The researchers are now working to identify the cell-surface receptors that attach to BRP. Receptors are molecular structures that receive signals from hormones, drugs, and other chemical messengers. Determining which receptor BRP uses will help scientists understand exactly how the peptide changes appetite and metabolism.

    The team also wants to map the full sequence of events that occurs after BRP binds to its target.

    Another challenge is duration. Small peptides are often broken down quickly in the body, which can shorten their effects. The researchers are investigating ways to make BRP last longer so that, if it eventually works in people, it could be administered on a more practical schedule.

    “The lack of effective drugs to treat obesity in humans has been a problem for decades,” Svensson said. “Nothing we’ve tested before has compared to semaglutide‘s ability to decrease appetite and body weight. We are very eager to learn if it is safe and effective in humans.”

    The longest-living family eats this 1 meal every day for lunch

    For secrets to living a long, healthy life, many people turn to the habits of people in the Blue Zones. These are areas in Italy, Greece, Japan, Costa Rica, the United States and more, where people regularly live into their 90s and 100s. 

    Bestselling author Dan Buettner has made it his life’s work to shine a light on these communities and understand their lifestyles, uncovering the habits that help them live longer than most of us.

    In a recent social media post, he shared the meal that “the longest-lived family in the world” eats every day.

    The Melis family, from Sardinia, Italy, holds a world record for longevity. They took the Guinness World Record title for the highest combined age for nine living siblings in 2012.

    “Nine siblings, collective age: 861 years. Oldest sibling is 109. Every day of their life, they had the exact same lunch,” Buettner said in the video.

    The 1 meal the longest living family eats everyday

    “The longest-lived family in the world eats this meal every single day,” Buettner wrote in the Instagram post. The meal consists of:

    • Three-bean minestrone (garbanzo, pinto, and white bean)
    • Sourdough bread
    • A glass of red wine (2-3 ounces)

    While many factors besides what you eat will play a role in how long you live, this meal exemplifies what Buettner calls the Blue Zones diet, which emphasizes whole foods and plants.

    Buettner said the family eats a chunky minestrone made with a variety of garden vegetables grown nearby, and it always contains three types of beans: garbanzo, pinto and white bean. 

    “People who live the longest, healthiest lives tend to eat half a cup to a cup of beans daily. Plus, the soup is rich in vegetables, which are top longevity foods,” Samantha Cassetty, a registered dietitian based in New York City and the co-author of “Sugar Shock,” tells TODAY.com.

    “It’s also noteworthy that the soup uses three types of beans and multiple vegetables. Research suggests that eating 30 unique plant foods per week can improve your gut diversity, a marker of a healthy gut. Your gut regulates health functions, like blood pressure and cholesterol, inflammation, mood, weight and nutrient absorption, so you’ll be more likely to live a longer and healthier life if you have a healthy gut,” Cassetty explains.

    Sourdough bread

    While bread often gets a bad rep, it can be part of a healthy diet —and choosing a sourdough variety can be especially beneficial for gut health.

    “People in the Blue Zones eat fermented foods daily. Naturally fermented sourdough bread … doesn’t produce the same blood-sugar spikes that white bread does, helping maintain healthier blood-sugar levels. Over time, high blood sugar can contribute to insulin resistance and type 2 diabetes,” says Cassetty. “Plus, there’s evidence that the resistant starch in sourdough bread may contribute to other health benefits, like increased satiety and better insulin sensitivity.”

    A glass of wine

    Buettner stressed that the family enjoys a “small” glass of wine, 2 or 3 ounces, half of a typical 5-ounce serving.

    “Alcohol can raise your good HDL cholesterol levels, but even a drink a day is associated with other health risks,” says Cassetty. “However, they may be drinking less than a drink a day. It’s also possible that wine boosts the enjoyment of their meal, and that’s a factor that often gets overlooked when we’re talking about healthy lifestyles.”

    And as Buettner pointed out, they didn’t choose this meal because “my diet requires me to do it.’ No, they loved it.”

    Cassetty adds: “Keep in mind that this meal is eaten in the presence of loved ones. Maintaining a strong social circle helps improve happiness and combat loneliness, bringing additional health benefits beyond the nutrients in the meal.”

    Is this a balanced, plant-based meal?

    A few commenters inquired about this lunch not containing enough protein, a common concern about plant-based meals. But this meal does deliver in the protein department.

    “A typical minestrone soup has 8 grams of protein per cup, but most people eat more than that,” says Cassetty. “A can of soup has closer to 2 cups, so you could get about 16 grams of plant-based protein from the soup. Sourdough bread can add another 4 grams of protein, bringing the total protein to roughly 20 grams.”

    “A balanced meal contains mostly whole, plant foods and adequate protein. This meal certainly fits the bill,” adds Cassetty. “You can get all the protein you need from plants as long as you’re meeting your calorie needs and varying your protein sources.”

    For those with higher protein needs, she recommends adding more beans to the soup, but also reminds us that it’s smart to spread your protein intake throughout the day rather than consuming all your protein in a single meal.

    Is eating the same meal every day a healthy choice? 

    Eating the same exact thing every day can sometimes be a red flag for dietitians, but Cassetty says this meal gets the green light.

    “I’d be concerned if someone was eating the same thing every day if it lacked plant diversity or was low in plant foods, but this meal is 100% plant-based and has a lot of variety within the meal,” says Cassetty. “Also, you can mix up the veggies in the soup using what’s in season, so that will add more plant variety to your diet.” 

    Eating the same midday meal can also help take the stress out of meal planning.

    “One thing people struggle with is making decisions about what to eat. This can be a real challenge when juggling family, work and other priorities,” says Cassetty. “Eating the same meal for lunch daily reduces decision fatigue, which can lower your stress levels. So, having the same meal on repeat can be an appropriate stress-busting routine, and reducing stress has health benefits and is a tenet of the Blue Zones philosophy.”

    What is the 321 Method for Ribs?

    Smoking ribs using the 321 method is one of the most popular ways for a backyard enthusiast to cook ribs. They result in a truly delectable, fall-off-the-bone rib. While many “BBQ purists” will balk at the 321 method, there’s no denying that this meat is juicy, tender, and delicious. So, if you or your guests like meat that comes clean off the bone with every bite, definitely give the 321 rib method a try!

    So what is the 321 method for ribs, exactly? It’s an easy way to cook pork ribs! In a nutshell, the 321 method for ribs is just what the name suggests:

    3 hours in the smoker

    Start by placing your ribs directly on the grill grates of the smoker for 3 hours to infuse the meat with that good, smoky flavor.

    2 hours wrapped

    Next up, wrap those smoky ribs in foil (usually with brown sugar, apple cider, butter, and other goodies) for 2 hours to add more flavor and keep that rib meat juicy.

    1 hour with sauce

    Last up, remove the meat from the wrapping and smother them in BBQ sauce. Finish them on the grill for 1 hour to set the sauce and ensure that the meat falls right off the bone.

    321 Rib Method

    This 321 rib method has gained a lot of popularity with home cooks in recent years because of the flawless results in churning out super tender ribs that fall off the bone every time.

    While competition and traditional BBQ cooks will scoff at fall-off-the-bone ribs, the truth is that most people really like them that way. Judges for KCBS won’t give you a winning score, but they also won’t be hanging out in your backyard critiquing what you and your family are eating. So I say, cook what you like!

    Sticky, sweet, tender, and oh-so-savory, 3 2 1 ribs are probably the most requested meal I receive for potlucks or family dinners. I tend to fall back on these ribs for gatherings because I know that they will deliver that tender, flavorful porky flavor every time. And sometimes consistency is super important!

    One final tip: I highly recommend reading my post on How to Know When Pork Ribs are Done. This will help you determine when to remove the ribs from the smoker in addition to the times suggested.

    INGREDIENTS

    How to Smoke 321 Ribs

    I firmly believe that you should cook food the way you like it, and ribs are no different! Do you like your ribs saucy? Put some extra sauce on them (I like using my Everything BBQ Sauce). Do you like your ribs falling off the bone? Follow this 321 rib method! These are your ribs after all. Make them exactly how you like them!

    Here’s how to cook your ribs using the 321 method:

    1. PREHEAT.
      • Fire up the grill and preheat to 180-200 degrees F. Prepare the ribs for smoking while the grill preheats.
    2. PREP THE RIBS.
      • Use a sharp knife to lift up the corner of the membrane on the back of the ribs. Using a paper towel, grip the membrane and pull firmly to fully remove the membrane. Trim any excess fat off the ribs as needed.
      • Next, season both sides of the ribs liberally with Sweet Rub or Rib Rub.
    3. SMOKE FOR 3 HOURS.
      • Start with exposing the seasoned ribs to 3 hours of heavy smoke over low heat. I prefer fruit woods (apple or cherry) when smoking pork, but other hardwoods (hickory and alder) also work really well with ribs.  
        • Place the seasoned ribs on the smoker and close the lid. Leave them to smoke for 3 hours (The ribs should be somewhere around an internal temperature of 165 degrees F at the end of these 3 hours).
    4. WRAP FOR 2 HOURS.
      • After the initial 3-hour smoke, foil the ribs with a liquid of your choosing (I used apple cider, dark brown sugar, and butter) and cook at 225-250 degrees F for 2 more hours. After the braising period, Iook for great retraction of the meat from the bones. You want the bones to be exposed on the end at least 1/4 to 1/2 an inch.
        • Tear off a large piece of aluminum foil and place it on a large working surface.
        • Transfer the ribs to the foil bone side up so the bones don’t tear through the foil.
        • Sprinkle the ribs with the brown sugar, top with the butter cut into small pads, and then pour in the apple cider.
        • Tightly crimp the foil together to create an airtight seal.
        • Return the wrapped ribs to the grill and increase the temperature in the smoker to 225 degrees F. Let the ribs braise for 2 hours in the foil.
    5. SAUCE AND SMOKE FOR 1 HOUR.
      • Lastly, baste the ribs in your favorite BBQ sauce (my Everything BBQ Sauce or Apple Jalapeno BBQ Sauce are delicious!)  and return them to the grill for a final hour to finish the ribs and set the sauce.
        • Carefully remove the ribs from the grill and place them on a large working surface.
        • Open the foil package (be careful of hot steam) and use tongs to remove the ribs and place them back on the grill, bone side down.
        • Discard the foil and excess cooking liquid. Next, brush the ribs with Everything BBQ Sauce (or your favorite BBQ sauce).
        • Return the unwrapped, sauced ribs to the smoker. Close the lid, and continue smoking for an hour (or less!) or until the ribs reach around 200 degrees F and the sauce is sticky and set.
    6. REST AND ENJOY.
      • Remove the ribs from the smoker and rest for 10-15 minutes. Slice into individual ribs and serve! I like to put extra sauce on the table for guests who enjoy their ribs extra saucy.
